ปัจจุบันเภสัชตำรับของไทยได้กำหนดมาตรฐานของขมิ้น
ซึ่งประกอบด้วย 5%
curcuminoid, 6% น้ำมันหอมระเหยสีเหลืองส้ม ประกอบด้วย monoterpenes,
sesquiter-penes, zingiberene, curcumene, a-turmerone
และ b-turmerone (1)
มีรายงานจากอินโดนีเซียว่าขมิ้นสามารถใช้รักษาอาการอุจจาระร่วงได้

ขมิ้นมีฤทธิ์คลายกล้ามเนื้อเรียบ
(4, 5) โดยออกฤทธิ์ต้าน acetylcholine, barium choride และ serotonin
(5) มีการทดลองในหนูขาวพบว่าขมิ้นมีฤทธิ์ลดการบีบตัวของลำไส้เล็กของหนูขาวได้
(6) ซึ่งกลไกการออกฤทธิ์เกี่ยวข้องกับฤทธิ์ต้านแคลเซียม (calcium
antagonism) (7) curcumin มีฤทธิ์ลดการบีบตัวของลำไส้สุนัข (8)
มีการทดลองทางคลินิกในคนไข้
440 คน อายุเฉลี่ย 48.5 ปี ทดลองโดยให้รับประทานขมิ้น
วันละ 162 มก. พบว่ามีฤทธิ์ลดการบีบตัวของลำไส้ได้ (9)
3. ฤทธิ์ต้านเชื้อแบคทีเรีย
สารสกัดขมิ้น
และน้ำมันหอมระเหยมีฤทธิ์ฆ่าเชื้อแบคทีเรียได้หลายชนิด
ทั้งแบคทีเรียที่เป็นสาเหตุของการแน่นจุกเสียด หรือท้องเสีย (10-16) ขมิ้นสามารถยับยั้งการเกิดกรดเนื่องจากเชื้อ
Lactobacillus acidophilus และ L.
plantarum (15) และยังออกฤทธิ์ยับยั้งการเกิดก๊าซเนื่องจากเชื้อ
Escherichia coli (16)
Monoterpene
และ sesquiterpene ในน้ำมันหอมระเหยจากใบ และ sesquiterpene
ketone ในน้ำมันหอมระเหยจากเหง้า สามารถยับยั้งเชื้อ Escherichia
coli MTCC 433 ได้ที่ความเข้มข้น 15.62 และ
125 มคล./มล. ตามลำดับ
(17) น้ำมันหอมระเหยในความเข้มข้น 1 M พบว่ามีฤทธิ์ต้านเชื้อ
Escherichia coli, Salmonella paratyphi, Salmonella typhosa และ Staphylococcus aureus (14) ความเข้มข้น 31.25
มคล./มล. มีฤทธิ์ต้านเชื้อ
Shigella flexnariและ Shigella sonnei ความเข้มข้น 62.5 มคล./มล. มีฤทธิ์ต้านเชื้อ Shigella boydii และความเข้มข้น 7.81 มคล./มล.
มีฤทธิ์ต้านเชื้อ Escherichia coli และ
Shigella dysnteriae (18) ในจานเพาะเชื้อ
นอกจากนี้ยังมีการศึกษาโดยทดลองใช้น้ำมันหอมระเหยของขมิ้นในค่าความเข้มข้นต่ำสุดที่สามารถยับยั้งเชื้อ
(MIC) เท่ากับ 125 มคก./มล. มีฤทธิ์ต้านเชื้อแบคทีเรีย (19) ในจานเพาะเชื้อ
และยังมีการทดลองใช้น้ำสกัด ความเข้มข้น 0.3 มล. มีฤทธิ์ต้านเชื้อ Escherichia coli, Salmonella
paratyphi, Salmonella typhi และ Staphylococcus
aureus (20)
น้ำสกัดและสารสกัดเฮกเซน
ความเข้มข้น 200 มก./มล. ไม่มีฤทธิ์ต้าน Escherichia
coli, Salmonella typhimurium และ Staphylococcus aureus (21)
ขณะที่น้ำสกัดและสารสกัดปิโตรเลียมอีเทอร์ ความเข้มข้น 250 มก./มล. มีฤทธิ์ต้านเชื้อ Escherichia

มีฤทธิ์ต้านเชื้อ Staphylococcus aureus (23) แต่ไม่มีฤทธิ์ต้านเชื้อ
Escherichia coli และที่ความเข้มข้น 200 มก./มล. ไม่มีฤทธิ์ต้านเชื้อ Escherichia
coli (21) เช่นกัน
สารสกัดเอทานอล
(95%) ความเข้มข้น 250 มก./มล.
ทดลองในอาหารเลี้ยงเชื้อ (24) และจานเพาะเลี้ยงเชื้อ
(22) พบว่ามีฤทธิ์ต้านเชื้อ Staphylococcus aureus
แต่ที่ความเข้มข้น 10 มก./มล. ไม่มีฤทธิ์ต้านเชื้อดังกล่าว (25)
สารสกัดอัลกอฮอล์และน้ำมันหอมระเหยของขมิ้นมีฤทธิ์ฆ่าเชื้อแบคทีเรีย Staphylococci (26) และน้ำมันหอมระเหยในขมิ้นกระตุ้นการเจริญเติบโตและสร้างกรดโดยเชื้อ
Escherichia coli ส่วน curcumin ไม่กดการเจริญเติบโตของแบคทีเรีย ยกเว้นเชื้อ Staphylococcus
aureus และในสารสกัดอัลกอฮอล์ของขมิ้นและน้ำมันหอมระเหยจากขมิ้นเหนี่ยวนำการเปลี่ยนแปลงโครงสร้างของ
Staphylococcus spp. (24) และน้ำมันจากขมิ้นที่สกัดด้วยเฮกเซนมีฤทธิ์ต้านเชื้อ
Staphylococcus aureus และ Escherichia
coli (27)
Curcumin
มีฤทธิ์ต้านเชื้อ E. coli (28), Staphyllococcus
spp. และ Streptococcus spp. แต่ไม่มีฤทธิ์ต้านเชื้อ
Proteus spp. และ Klebsiella spp.
ในจานเพาะเชื้อ (29) curcumin ยังมีฤทธิ์ต้านเชื้อ
S. aureus แต่ไม่มีฤทธิ์ต้านเชื้อ Lactobacillus
acidophilus และ Salmonella typhosa ในหลอดอาหารเลี้ยงเชื้อ (30)
นอกจากนี้ยังมีการทดลองโดยใช้
curcumin ความเข้มข้น 1 ไมโครโมล พบว่ามีฤทธิ์ต้านเชื้อ S.
aureus และความเข้มข้น 10 ไมโครโมล
มีฤทธิ์ต้านเชื้อ Salmonella typhimurium และ E.
coli ในจานเพาะเชื้อ

มีการทดลองพบว่าสารสกัดด้วยอัลกอฮอล์และน้ำ
(1:1) ความเข้มข้น 125 มคก./ซีซี
มีผลในการฆ่าเชื้ออะมีบา (Entamoeba histolytica) ซึ่งเชื้อนี้เป็นสาเหตุของโรคบิดมีตัว

1.
การทดสอบความเป็นพิษ
เมื่อฉีดสารสกัดเอทานอล
(95%) ให้แก่หนูถีบจักรทางช่องท้อง
พบว่าขนาดที่ให้หนูตายครึ่งหนึ่ง (LD50) คือ 3.98 ก./กก. ส่วนการฉีดสารสกัดจากน้ำและปิโตรเลียมอีเทอร์เข้าทางช่องท้องหนูถีบจักร
พบว่า LD50 มีค่า 430 มก./กก. และ 525 มก./กก. ตามลำดับ (33) เมื่อฉีดสารสกัดจากเอทานอลและน้ำ (1:1) ให้แก่หนูถีบจักรทางช่องท้อง พบว่า LD50
มีค่าเท่ากับ 500 มก./กก. (32)
เมื่อกรอกสารสกัดจากเอทานอล
(95%) ให้หนูถีบจักร ในขนาด 100 มก./กก./วัน
ไม่มีผลต่อน้ำหนักตัว แต่มีผลต่อปอด หัวใจ เม็ดเลือดแดง
และเม็ดเลือดขาวลดลง แต่ไม่มีผลต่ออสุจิ (34) การศึกษาพิษกึ่งเรื้อรังในหนูถีบจักรและหนูขาว
โดยให้ขมิ้น 0, 1, 5% และสารสกัดเอทานอล (95%) โดยผสมในอาหารให้หนูกิน 14 และ/หรือ 90 วัน พบว่าผงขมิ้นในขนาดสูง (5%) และในระยะเวลาที่นาน (90 วัน) พบว่าน้ำหนักลด
เป็นพิษต่อตับ มี necrosis ในหนูถีบจักรในขนาดน้อย 0.2 หรือ 1% เป็นเวลา 14 วัน พบพิษต่อตับ
พิษต่อหนูถีบจักรมากกว่าหนูขาว ส่วนสารสกัดในขนาดที่ให้ไม่พบพิษ (35)
การทดลองในคนทั้งชาย-หญิง โดยให้รับประทานขมิ้นในขนาดวันละ 2.2 ก.
ไม่ทำให้เกิดพิษ (36)
เมื่อผสมสารสกัดเอทานอล
(95%) ให้แก่หนูขาวทั้งเพศผู้ และลิง เพศผู้-เมีย ในขนาด 300 มก./กก.
หนูตะเภาเพศผู้ ขนาด 2.5 ก./กก. และหนูขาวเพศผู้-เมีย
ในขนาด 2.5 ก./กก. ไม่ทำให้เกิดพิษ (37) กรอกสารสกัดอัลกอฮอล์และน้ำ (1:1) ให้หนูถีบจักร ในขนาด 4 มก./กก. ไม่มีผลต่อการกินอาหาร
การเพิ่มของน้ำหนัก หรือระบบประสาทส่วนกลาง (38) ผสม oleoresin จากขมิ้นในอาหารให้แก่หมูในขนาด
60, 296, 1551 มก./กก./วัน เป็นเวลา 102-109
วัน พบว่าทำให้เกิด thyroid hyperplasia เกิดการเปลี่ยนแปลงของ epithelial
ของกระเพาะปัสสาวะ ไต และ perichalangitis (39) ผสม oleoresin ลงในอาหารให้แก่หนูขาวเพศผู้และเพศเมียในขนาด
0, 1,000, 5,000, 10,000, 25,000 และ 25,000 ppm พบว่าไม่มีการเปลี่ยนแปลงอย่างมีนัยสำคัญของส่วนประกอบของเลือด แต่จะเกิด hyperplasia
ของ mucosal epithelium ใน cecum และ colon ในหนูที่ได้รับ oleoresin 50,000
ppm (40)
ผงขมิ้นไม่ก่อให้เกิดพิษเฉียบพลัน
เมื่อให้หนูถีบจักรในขนาด 10 ก./กก. ส่วนสารสกัดเอทานอลเมื่อให้หนูถีบจักรโดยการฉีดเข้าทางช่องท้อง
ทางใต้ผิวหนัง หรือให้กิน มีค่า LD50 มากกว่า 15 ก./กก.(6) ส่วนการทดสอบพิษกึ่งเรื้อรังโดยให้ในขนาด 0.03,
2.5 และ 5 ก./กก./วัน แก่หนูขาว 96 ตัว เป็นเวลา 6 เดือน พบว่าหนูเพศผู้ที่ได้รับสาร (ผงขมิ้น) 2.5 และ 5 ก./กก./วัน กินอาหารได้น้อยลง
และอัตราการเจริญเติบโตน้อยลง เมื่อเทียบกับกลุ่มควบคุม (41)
การทดสอบพิษเฉียบพลันพบว่าไม่เกิดพิษเฉียบพลันในหนูขาว
หนูตะเภา และลิง เมื่อให้ขมิ้นในขนาด 2 ก./กก. หรือสารสกัดจากอัลกอฮอล์ของขมิ้นในขนาด 300 มก./กก. (37)
2.
ฤทธิ์ก่อกลายพันธุ์
สารสกัดเอทานอล (95%) ในขนาด 10 มก./จานเพาะเชื้อ มีฤทธิ์ก่อกลายพันธุ์ต่อ Salmonella
typhimurium TA102 (42) สารสกัดเหง้าขมิ้นด้วยเอทานอล (95%)
ในขนาด 15 มก./จานเพาะเชื้อ มีฤทธิ์ก่อกลายพันธุ์ต่อ S.
typhimurium TA98 (43) เมื่อฉีดสารสกัดจากน้ำเข้าท้องหนูถีบจักร
ขนาด 40 มก./จานเพาะเชื้อ
ไม่มีฤทธิ์ก่อกลายพันธุ์ต่อ S. typhimurium TA100 และ
TA 98 (44) สารสกัดขมิ้นด้วยน้ำในขนาด 100 มก./จานเพาะเชื้อ ไม่มีฤทธิ์ก่อกลายพันธุ์ต่อ Cells-Pig-Kidney-LLC-PK-1 และ Cells-trophoblastic placenta (45) สารสกัดขมิ้นด้วยน้ำ
ขนาด 50 มก./ซีซี
ไม่มีฤทธิ์ก่อกลายพันธุ์ใน S. typhimurium TA1535 (46) สารสกัดขมิ้นด้วยน้ำหรือน้ำร้อน หรือผงขมิ้น ขนาด 0.5 ซีซี/แผ่น ไม่มีฤทธิ์ก่อกลายพันธุ์ต่อ Bacillus
subtilis H-17 (Res+, Res-) และ H-45
(Res-) (47) สารสกัดด้วยน้ำร้อนและอัลกอฮอล์ ขนาด 50
มก./แผ่น (dry weight of plant) ไม่มีฤทธิ์ก่อกลายพันธุ์ต่อ
S. typhimurium TA98 และ TA100 (48) สารสกัดจากเอทานอล (95%) ในขนาด 250 และ 360 มคก./จานเพาะเชื้อ ไม่มีฤทธิ์ก่อกลายพันธุ์ต่อ S.
typhimurium TA98, TA100 และ TA1535 (49) เมื่อใช้ขมิ้นจากส่วน
bulb ในขนาด 50 มคก./จานเพาะเชื้อ
ไม่มีฤทธิ์ก่อกลายพันธุ์ต่อ S. typhimurium TA1537 และ TA1538 (50) Resin ในขนาด 160 มคก./จานเพาะเชื้อ ไม่มีฤทธิ์ก่อกลายพันธุ์ต่อ S. typhimurium TA98,
TA100 และ TA1535 (51) เมื่อให้ขมิ้น 0.5% หรือ curcumin 0.015% ผสมในอาหารให้หนูถีบจักร
พบว่าไม่ทำให้โครงสร้างและจำนวนของโครโมโซมของไขกระดูกเปลี่ยนแปลงและไม่ก่อกลายพันธุ์
และเมื่อให้หนูขาวกินอาหารผสมขมิ้น 0.5 และ 0.05% ซึ่งทำให้สุกไม่พบพิษต่อโครโมโซม (52) Sodium curcuminate เป็นเกลือที่ได้จากผงขมิ้นทำให้เกิดความผิดปกติของโครโมโซมในตั๊กแตน และ Poecilocera picta
(Acrididae) (53) สารสกัดหยาบของขมิ้นไม่มีฤทธิ์ก่อกลายพันธุ์ เมื่อทดสอบโดย Ames test โดยใช้ Salmonella typhimurium TA98 และ TA100
(54) สารสกัดขมิ้นด้วยเอทานอล(95%) ไม่มีฤทธิ์ก่อกลายพันธุ์ใน
S. typhimurium T1530 (55)
Curcumin ไม่มีฤทธิ์ก่อกลายพันธุ์ (55) ขมิ้นชันมีฤทธิ์ก่อกลายพันธุ์
(42, 57) แต่มีรายงานว่าสารสกัดขมิ้นชันไม่มีฤทธิ์ก่อกลายพันธุ์ใน
S. typhimurium TA98 และ TA100 (58)
3.
ทำให้ตัวอ่อนผิดปกติ
เมื่อใช้ส่วนรากผสมลงไปในอาหาร
(0.5%) ให้หนูขาวเพศเมีย พบว่าไม่มีฤทธิ์ teratogenic
(52) เมื่อป้อนสารสกัดเอทานอล
(95%) สารสกัดจากน้ำและสารสกัดปิโตรเลียมอีเทอร์ ในขนาด 200 มก./กก.ให้แก่หนูขาว
หรือกระต่ายเพศเมีย ไม่พบฤทธิ์ teratogenic (59)
4.
พิษต่อตัวอ่อน
เมื่อให้หนูขาวเพศเมียกินสารสกัดจากเอทานอล
(95%) สารสกัดจากน้ำและสารสกัดจากปิโตรเลียมอีเทอร์ ในขนาด 100 มก./กก. พบว่าเป็นพิษต่อตัวอ่อน
(60) เมื่อให้หนูขาวเพศเมียกินสารสกัดจากน้ำและสารสกัดจากปิโตรเลียมอีเทอร์ในขนาด
200 มก./กก. พบว่ามีพิษต่อตัวอ่อน
และในขนาด 100 มก./กก. ไม่มีพิษ ส่วนสารสกัดเอทานอล (95%) ในขนาด 200 มก./กก. และ 100 มก./กก. ไม่มีพิษต่อตัวอ่อน (61)
5.
พิษต่อยีน
เมื่อกรอกผงขมิ้นให้แก่หนูถีบจักร ในขนาด 2.5, 5 และ
7.5 ก./กก. ไม่พบว่ามีพิษ

6.การทดสอบความเป็นพิษของโซเดียมเคอร์คิวมิเนท ซึ่งเป็นสารสำคัญของขมิ้น
พบว่าในขนาด 500 มก. เมื่อให้ทางปาก ฉีดเข้าใต้ผิวหนัง
ฉีดเข้าช่องท้อง ไม่พบพิษในหนูขาว
แต่ในขนาดเดียวกันเมื่อฉีดเข้าหลอดเลือดหนูถีบจักร พบว่าสัตว์ทดลองตายหมด ในขนาด 250
มก./กก. ตาย 3 ใน 6 และ 100 มก./กก. ไม่พบพิษ (63)
7. ไม่ควรใช้ในผู้ป่วยที่มีปัญหาเรื่องท่อน้ำดีอุดตัน
หรือผู้ที่เป็นนิ่วในถุงน้ำดี ต้องอยู่ในความดูแลของแพทย์ (64)
8. ไม่ควรใช้ในหญิงมีครรภ์ (64)
9. การผสมเพิ่มในอาหารหนูจนถึง 10% ไม่มีผลทำให้เคมีเลือดเปลี่ยนแปลง (65, 66)
10. Oral LD50 ของเคอร์คิวมินในหนูถีบจักร มีค่ามากกว่า 2 ก.ก/กก. (67)
11. ผงขมิ้นทำให้มีผู้แพ้
(68) และสารสกัดขมิ้นในขนาด 1% ทำให้แพ้ (69)
จากผลการทดลองข้างต้นจะเห็นว่าขมิ้นสามารถใช้รักษาอาการท้องเสียได้
โดยฆ่าเชื้อแบคทีเรียที่เป็นสาเหตุของท้องเสียได้หลายชนิดและยังลดการบีบตัวของลำไส้ และค่อนข้างปลอดภัย
แต่เนื่องจากผู้ป่วยเอดส์มักมีอาการติดเชื้อรุนแรง ขมิ้นอาจไม่ทันกับโรค
